RESUMEN

Objective: To investigate and monitor the occupational hazards in the Teaching and Research Laboratory (hereinafter referred to as the place) of a university, so as to provide basis for the occupational health work in the university. Methods: November 2014, 46 places in a university were selected by stratified random sampling, and the occupational health risk factors were investigated. Results: Indoor temperature, humidity, sulfur dioxide, nitrogen dioxide, carbon monoxide and carbon dioxide were detected in 21 sites, xylene and hydrofluoric acid were detected in 6 sites, and colony count was detected in 18 sites, the power frequency electric field intensity was measured in 23 places, and the x-ray radiation dose was measured in 4 places. Noise was measured at 21 sites, with 7 sites exceeding the standards accounting for 33.3% (7/21) ; 21 sites were detected for illumination and 10 sites for nonconformity accounting for 47.6% (10/21) ; 10 sites for Microwave Radiation and 3 sites exceeding the standards accounting for 30% (3/10) ; and 25 sites were detected for outdoor air volume and air velocity, the percentage of unqualified was 72% (18/25) in 18 sites, among which the wind velocity was statistically significant in teaching, research and experimental sites (P=0.010) . Conclusion: The occupational hazards in the teaching and research places of a university should be paid attention to, and the engineering protection and personal protection should be strengthened in the experiment.

RESUMEN

Objective Recently, the incidence of breast cancer has been the highest among female malignant tumors. Therefore, potential biomarkers are urgently needed to predict and prevent breast cancer. This study was aimed to explore the expression and clinical significance of differential miRNAs and their target genes in breast cancer by establishing miRNA expression profile in breast cancer tissues. Methods From January 2015 to December 2018, a total of 137 cases of breast cancer tissues with paired paracancerous tissues and 20 cases of breast fibroadenoma tissues were collected from the department of breast surgery, affiliated hospital of Guizhou Medical University. The tissues were divided into breast cancer group, paracancerous group, breast fibroadenoma group and lymph node metastasis group. High-throughput sequencing technique was used to detect the miRNAs in breast cancer tissues and paired paracancerous tissues. Real-time PCR verified the expressions of the three miRNAs with the most significant expression differences in different groups. Finally, bioinformatics analysis was used to predict the target genes and investigate the expression of miRNAs and proteins of target genes in different breast diseases. Results A total of 157 upregulated and 162 downregulated miRNAs were screened by high-throughput sequencing. Mir-hsa-miR-532-3p and hsa-miR-1260b were the most significant in upregulated miRNAs while has-let-7c-5p was the most significant in downregulated miRNAs. Meanwhile, bioinformatics analysis showed that their target gene was EZH2. Compared with para-cancerous group, expressions of hsa-miR-532-3p and hsa-miR-1260b were significantly upregulated while hsa-let-7c-5p was significantly down-regulated in breast cancer group and lymph node metastasis group (all P<0.05). The miRNA expressions of target gene EZH2 in breast cancer group, breast fibroadenoma group and lymph node metastasis group (1.24±0.01, 4.02±0.01, 15.97±0.01, respectively) were upregulated when compared with the para-cancerous group (1.00±0.00), and the similar conclusion could be drawn in EZH2 protein expression. Conclusion Hsa-miR-532-3p, hsa-miR-1260b and hsa-let-7c-5p were closely related to breast cancer, which may promote the occurrence and development of breast cancer by inducing the transcriptional expression of EZH2. HsamiR-532-3p, hsa-miR-1260b, hsa-let-7c-5p and EZH2 may be potential tumor biomarkers of breast cancer.

RESUMEN

PURPOSE: ORM1-like 3 (ORMDL3) belongs to a highly conserved protein family which is anchored as transmembrane protein in the endoplasmic reticulum. Gasdermin B (GSDMB) is adjacent to ORMDL3 on chromosome 17q21.2 and belongs to the gasdermin-domain containing the protein family (GSDM family). Recent reports suggest that GSDMB and ORMDL3 are associated with asthma in several populations. However, genetic association studies that examined the association of GSDMB and ORMDL3 gene variants with asthma showed conflicting results. To assess whether combined evidence shows the association between GSDMB/ORMDL3 polymorphism and asthma. METHODS: A bibliographic search from MEDLINE identified 13 original articles using the search keywords 'GSDMB', 'ORMDL3', and 'asthma'. An updated literature-based meta-analysis involving 6,691 subjects with asthma, 9,281 control individuals, and 1,360 families were conducted. Meta-odds ratios (ORs) and 95% confidence intervals (CIs) based on the fixed effects model or the random effects model depended on Cochran's Q-statistic and I2 values. Data from case-control and TDT studies were analyzed in an allelic model using the Catmap software. RESULTS: We selected and identified 3 SNPs of ORMDL3 associated with asthma (rs8076131: OR=1.10; 95% CI, 1.02-1.20; P=0.012. rs12603332: OR=1.15; 95% CI, 1.05-1.25; P=0.002. rs3744246: OR=1.10; 95% CI, 1.02-1.17; P=0.008) and 1 SNP of GSDMB associated with asthma (rs7216389: OR=1.37; 95% CI, 1.27-1.47; P<0.01). Publication bias was estimated using modified Egger's linear regression test proposed by Harbordetal and revealed no evidence of biases. Furthermore, cumulative meta-analysis in chronological order showed the inclination toward significant association for rs7216389 and rs12603332 with continually adding studies, and the inclination toward null-significant association for rs3744246 and rs8076131. CONCLUSIONS: Moderate evidence exists for associations of the ORMDL3 rs8076131, rs12603332, and rs3744246 and GSDMB rs7216389 variants with asthma. Large sample size and representative population-based studies and TDT studies with homogeneous asthmatic patients and well-matched controls are warranted to confirm this finding.

RESUMEN

<p><b>OBJECTIVE</b>Effects of inter-individual variability on fMRI of acupuncture were observed and the possible influencing factors were further analyzed.</p><p><b>METHODS</b>Twenty-six healthy volunteers were selected. And acupuncture was applied at Zusanli (ST 36) on the left side with even manipulation. The same experimental designation and data collecting reference were adopted to collect functional data. Then, the same data processing method was applied for analyzing individual data. Data which did not confirm with data analyzing qualification were rejected. The 26 individual data which met the requirement were taken randomly for 5 times according to the principle of random group division. Five groups named with A, B, C, D and E were thus generated with 11 samples in each. Images were processed with the AFNI software for every group, and the activated brain areas were revealed.</p><p><b>RESULTS</b>Activated areas in the brain were observed in all the 5 groups, and the results vary a lot among different groups. Decreased signals of activated brain areas were observed in group C, while increased signals were seen in group D. Partial increasing and partial decreasing signals appeared in the other 3 groups. Compared with other groups, group D demonstrated totally different activated areas. The rate of difference among different groups is 46.7%-100.0%, and most of the differences were over half of the activated areas.</p><p><b>CONCLUSION</b>Under the pre-requisites of strict control of experimental designation, acupuncture method, data collecting and processing, great differences have been found in the activated areas of the brain. It indicates that obvious individual differences existes in the activated areas of the brain with acupuncture. And the difference may greatly influence the researching result of fMRI as well as conclusions of those results.</p>

<p><b>OBJECTIVE</b>To study the changes of pulmonary function in children with right lung middle lobe syndrome before and after treatment.</p><p><b>METHODS</b>Thirty children with right lung middle lobe syndrome were classified into two age groups: < or =4 years old and >4 years old. Pulmonary function was tested by the 2600-type and the MIR-type pulmonary function spirometry in the < or =4 years and the >4 years age groups, respectively before and after treatment. Terminal flows/peak expiratory flow (25/PF) and the percentage of tidal volume to peak tidal expiratory flow (% V-PF) were measured in the <4 years age group. Forced vital capacity (FVC), forced expiratory volume in one second (FEV1) and peak expiratory flow (PEF) were measured in the >4 years age group.</p><p><b>RESULTS</b>The values of 25/PF and %V-PF in the < or =4 years age group were 0.42+/-0.08 and 0.28+/-0.03, respectively before treatment. The values were improved after treatment (0.58+/-0.12 and 0.39+/-0.06 respectively) (P<0.05). The values of FVC, FEV1 and PEF were 1.75+/-0.32, 1.36+/-0.52 and 2.56+/-0.78, respectively in the >4 years age group before treatment. The values were also improved after treatment (2.37+/-0.78, 2.08+/-0.65 and 3.68+/-0.80 respectively) (P<0.05).</p><p><b>CONCLUSIONS</b>There are significant differences in the pulmonary function before and after treatment in children with right lung middle lobe syndrome. The pulmonary function can return to normal after treatment.</p>

In combination with anatomy, record of ancient literature and teaching experience, the location of Naohui (TE 13), Chengshan (BL 57), Fengshi (GB 31), Zhongdu (GB 32), Toulinqi (GB 15), Yinbao (LR 9) and Shaoshang (LU 11) in the national standard are analyzed and the relative location methods are raised.

<p><b>OBJECTIVE</b>To investigate whether there is a possible role of pro-inflammatory cytokine high mobility group box protein 1 (HMGB1) causing liver failure in severe hepatitis B patients.</p><p><b>METHODS</b>Serum HMGB1 levels of chronic hepatitis B (CHB) patients with different clinical conditions were measured and the correlations between HMGB1 and TBil or PTA were analyzed. (1) 54 chronic hepatitis B patients in different clinical conditions were enrolled in our study. Their serum TBil and PTA levels were detected by routine methods. (2) Their serum HMGB1 levels were also detected. 100 KD super-filtration columns were used to get rid of large proteins in the serum and 10 KD columns were used to condense the protein. Western blot was used to determine HMGB1 levels, and correlations between HMGB1 and TBil or PTA were analyzed.</p><p><b>RESULTS</b>The detection rates of serum HMGB1 were 100% (23/23), 90% (9/10), and 55% (6/11) in 23 patients with hepatic failure, 10 patients with chronic severe hepatitis B, and 11 patients with chronic moderate hepatitis B respectively. The concentration of serum HMGB1 levels in these three groups was (83.4+/-21.3), (78.1+/-19.5) and (60.3+/-14.3) microg/L respectively. Serum HMGB1 was not detected in normal healthy controls and hardly detected in convalescent and mild hepatitis patients. There were positive correlations between HMGB1 and TBil and negative correlations between HMGB1 and PTA.</p><p><b>CONCLUSION</b>HMGB1 levels in serum were closely associated with disease severity in chronic hepatitis B patients. HMGB1 may play a key role in the pathogenesis of chronic severe hepatitis B and liver failure.</p>

UNLABELLED@#OBJECTIVE; To determine the effect of a variation of CAG-rich region, which was found in the 5'-regulatory sequence of insulin receptor substrate-1(IRS-1) gene in Type 2 diabetes mellitus(T2DM) patients, on gene expression and its mechanism.@*METHODS@#The recombinants, pGL2.P-T3 and pGL2.P-T5, were constructed with luciferase reporter vector, pGL2 promoter. T3 and T5 were wild-type and variant alleles, respectively. The recombinants were cotransfected with pSV-beta-galactosidase control vector to Hela cells. Luciferase assay was performed to assess transcriptional activity. The electrophoresis mobility shift assay(EMSA) and DNA footprint assay were applied to determine the interaction between the DNA regulatory sequences and nuclear proteins of Hela cells.@*RESULTS@#The relative transcription activity of T5 was lower than that of T3 [(7.76+/-1.05)% vs (9.98+/-1.40)%, P<0.05]; EMSA showed both T3 and T5 formed a single retarded band in gel with the same mobility with nuclear proteins; T5 had 2 binding sites for transacting factors, CGCGCCCGCGGGCGGCGGC and GGGCGGCTGGTGGCGGCTG, which was the same as T3.@*CONCLUSION@#Although the variation in T5 do not alter the DNA-binding sites for Hela cell nuclear extracts, the notable decrease in gene transcription activity induced by it may be an important factor to the development T2DM in the carrier.

<p><b>OBJECTIVE</b>To study on effects of acupuncture at a same point at physiological or pathological state on brain functional imaging.</p><p><b>METHODS</b>Electroacupuncture was given at Neiguan (PC 6) of the healthy senile persons (normal group) and the patients of Alzheimer's disease (AD group), and they were scanned on a 1.5 T MRI scanner and the data of functional magnetic resonance imaging (fMRI) were analyzed by Statistical Parametric Mapping (SPM99). The functional imaging of different brain regions were obtained from both the healthy group and the AD group.</p><p><b>RESULTS</b>Both the frontal lobe and the temporal lobe were activated in varying degrees by electroacupuncture at Neiguan (PC 6) in the healthy group. However, the frontal lobe, the temporal lobe and the cingulate gyrus as well as the cerebellum were activated in varying degrees and other brain regions were not activated by electroacupuncture at Neiguan (PC 6) in the AD group.</p><p><b>CONCLUSION</b>Acupuncture at Neiguan (PC 6) can activate functions of different brain regions in the healthy group and the AD group, indicating that acupuncture at Neiguan (PC 6) has different effects on functional magnetic resonance imaging in different functional states.</p>

RESUMEN

Objective To study the clinic effects and safety of budesonide suspension combined with tubertaline solution for nebulization therapy on infantile asthma.Methods Seventy-five cases with asthma were divided into 3 groups randomly,double-blinded and controlled,each group having 25 cases.Group A inhaling tubertaline solution(bricanyl),while group B inhaling pulmicort respules combined with bricanyl,group C inhaling budesonide suspension(pulmicort respules)only.Results In group A,16 cases was cure,3 cases was markedly improved,6 cases was failure,the total efficacy rate was 76 percent.In group B,22 cases was cure,2 cases was markedly improved,1 case failed,the total efficacy rate was 96 percent.While in group C,14 cases was cure,4 cases was markedly improved,7 cases failed,the total efficacy rate was 72 percent.The difference of efficacy rate between group A and group B was significant(P0.05).Conclusion Pulmicort respules combined with bricanyl for nebulising inhalation is effective and safety on infantile asthma and its effect is better than inhaling bude-sonide or tubertaline only.
